Jonathan Z. Sexton PhD.
Thesis title: High Content Assay Development for Glucose Stimulated Insulin Release: Discovery of Small Molecule Incretin Mimetics
The hypothesis/focus of this research was to develop new therapeutic incretin small molecules through high content screening (HCS) assay for GSIS for the treatment of T2DM.
Aims of this project were to validate the INS-1 preproinsulin mCherry cell line for GSIS, to develop and validate a pair of end point assays that can be used for insulin vesicle trafficking and to screen for small molecule incretins, to evaluate various constructs for high content screening using different fluorescent protein constructs, also to generate a stable cell line and pilot screening for small molecule incretins. The central dogma of insulin was looked at as well as the effects of GLP-1 on the different populations of insulin on the cell and other small oral bioavailable incretin drugs are screened
